How to convince my boss to promote me?

I was a Projects Engineer in a well known organization then I decided to switch career and work in quality assurance systems design and implementation. The new responsibility in my new job deserves a promotion. However HR in my company claims it doesn’t! I don’t know whether I should stay in my job unsatisfied with the grade and salary but hoping to get a chance for regional position as promised by my dotted line boss. Or leave the company whilst I don’t yet have another option! Need advice
